Basic Properties

1. Physical Properties


Appearance: Colorless, transparent liquid (high purity)


Concentration Range: Common commercial solutions are 30%, 40%, and 50% (w/v)


Density: Approximately 1.07–1.12 g/cm³ (increases with increasing concentration)


Viscosity: Low viscosity (approximately 2–5 cP for 30% solution, 25°C)


pH (Neutral Conditions): 5.0–7.0 (unbuffered)


Solubility: Completely miscible with water, ethanol, and acetone


2. Chemical Stability and Reactivity


Polymerization: Easily undergoes free radical polymerization under light, heat, or ammonium persulfate (APS) + TEMED initiation, producing polyacrylamide (non-toxic gel).


Hydrolytic Stability: Slowly hydrolyzes to acrylic acid under acidic or alkaline conditions.


Storage Stability: Stable for 6–12 months under light-protected, refrigerated at 4°C, and neutral pH conditions.


Main Applications

1. Polyacrylamide Gel Electrophoresis (PAGE)

The most common application is in molecular biology for protein and nucleic acid separation.

  • SDS-PAGE: Denaturing protein gel electrophoresis.

  • Native PAGE: Preserving protein structure during separation.

  • DNA sequencing gels: High-resolution acrylamide gels for fragment analysis.

2. Manufacturing of Polyacrylamide Polymers

Acrylamide solution is polymerized to produce polyacrylamide, which serves as:

  • Flocculants for wastewater treatment.

  • Thickeners in enhanced oil recovery (EOR).

  • Soil conditioners in agriculture.

  • Paper strength additives in pulp and paper industry.

3. Crosslinking Agent in Hydrogels

When combined with bis-acrylamide, it forms crosslinked hydrogels used in:

  • Tissue engineering scaffolds.

  • Contact lenses (historical use; now mostly replaced).

4. Laboratory Reagent for Molecular Cloning

  • Protein crystallization studies.

  • Western blotting gel preparation.

  • Capillary electrophoresis sieving matrices.

Acrylamide-based copolymers improve adhesion, water resistance, and film formation in:
